REFP-1型电化学发光成像分析仪

在临床诊断检测和病理研究等生命科学相关领域研究中,“可视化”技术和相关仪器的开发越来越受到人们的重视。电化学发光成像技术是一种基于电化学发光反应进行的成像技术,不仅能够提供电化学信号和发光强度的信息,而且能够对其进行光学成像分析。作为一种新的生化分析手段,电化学发光成像具有设备简单、反应可控、多信息化、可视化、高灵敏度和高通量等优点。相对于荧光成像法,具有无光漂白,不需要光源和分光系统的优点;相对于化学发光成像法,具有可控性强的优点;相对于电化学检测,具有检测限低和受电极污染影响小的优点。由于在电化学发光生物传感成像中,标记物被固定在电极表面,所以不存在扩散影响成像分辨率的问题,量子产率高,响应时间短,稳定性好,可以用于高灵敏度电化学发光成像分析。本公司新进研发的REFP-1型电化学发光成像分析仪采用EMCCD图像传感器,对环境的要求低、操作简单方便、摄像时间短、灵敏度高,并且可以获得多个影像并存储。为了进一步提高发光检测灵敏度,仪器还设计有可与成像系统位置互换的PMT检测系统,使其能够完成灵敏度更高的检测需求。基于电化学发光成像高灵敏、反应可控、快速成像以及对于生物分析兼容性的特点,本仪器可以应用于潜在指纹的显现、成分识别、DNA检测、兴奋剂检测、病患临床诊断以及爆炸物检测等领域,具有重要的医学诊断、病理研究和安全保障价值。生物方面可以应用于免疫分析、基因表达、组织切片和单细胞检测、离体器官灌流、动物体内检测、肿瘤研究、细胞移植、传染性疾病、细胞凋亡、蛋白与蛋白之间的相互作用。将其发展为一种集成像分析、生物分析、指纹识别于一体的前瞻研究,为电化学发光成像技术在生命科学和法庭科学中的应用提供新的方法和思路。

REFP-1型电化学发光成像分析仪由陕西师范大学与西安瑞迈分析仪器有限责任公司合作研制开发。该仪器能完成在一份样品溶液中进行多组分测量,节省样品溶液,提供更多定性定量信息。

REFP-1型电化学发光成像分析仪由检测系统、控制测量系统构成

* 检测系统主要包含PMT组件、EMCCD成像检测组件、精密移动平台,可将被测样品精确移动到相应测试区域内,分别实现成像和发光的检测。

* 控制系统主要包含电化学测量、发光测量、移动平台和多电极电位控制,接受来自于计算机系统的控制指令,实现对被测样品的PMT检测、EMCCD成像和精确控制检测位置,实现对整个系统的数据采集、图像显示。

REFP-1 electrochemical luminescence imaging analyzer

"Visualization" technology and related equipment development more and more pay the attention in clinical diagnostic tests and pathological studies of the related fields in life science research. Electrochemical luminescence imaging technology is a kind of imaging technology based on the electrochemical luminescence response, and it not only can provide information on electrochemical signals and luminescence intensity, but also can analysis the electrochemical optical imaging. As a new biochemical analysis method, electrochemical luminescence imaging device have the characteristic of simple, reaction control, informationization, visualization, high sensitivity and high-flux. Compared with the fluorescence imaging method, it has without light bleaching, light source and spectral system; compared with the chemiluminescence imaging method, it have the advantage of strong controllability; Relative to the electrochemical detection, it have the advantages of low detection limit and less of the electrode pollution. Due to the electrochemical luminescence, markers are fixed on the electrode surface in biological sensing imaging, so there is no diffusion problem to affect the resolution of imaging quantity. The instrument is high quantum yield, short response time, good stability, and it can be used for high sensitive electrochemical luminescence imaging analysis.

Our company research and development of REFP-1 electrochemical luminescence imaging analyzer used EMCCD image sensor, the requirement of environment is low, operation simple and convenient, camera time is short, high sensitivity, and it can obtain multiple images and stored. In order to further improve the detection sensitivity, instrument designed PMT detecting system, which can interchange with the location of imaging system, so it can finish higher sensitivity testing requirements. Based on the electrochemical luminescence, high sensitive, reaction control, fast imaging and the analysis for biological compatibility, the instrument can be applied to potential fingerprint appeared, composition identification, DNA testing, doping tests, clinical diagnosis, explosives detection and other fields, it has important medical diagnosis, pathological study and security value. In biology aspect, it can be applied to the immune analysis, gene expression, tissue section and single cell test, in vitro organ perfusion, animal testing, cancer research, cell transplantation, infectious disease, apoptosis, interaction between protein and protein. It integrated of imaging analysis, biological analysis and fingerprint identification, and it can provide a new method and train of thought for the electrochemical luminescence imaging technology in life science and the application of forensic science.

Composition

REFP-1 electrochemical luminescence imaging analyzer consists of detection system and the control system:

* Test system mainly consists of PMT components, EMCCD imaging detection components, precision mobile platforms. Samples can be measured accurately moved to the corresponding test area, respectively, for detecting imaging and lighting.

* Control system mainly includes electrochemical measurement, luminescence measurement, mobile platforms and multiple electrode potential control, and it accept from computer system to control instruction, to control precise position of the detected PMT and EMCCD imaging, and to make sure entire system of data acquisition, image display.
